Cool Water hose leak and air in system ?

I just replaced water pump and thermostat and some hoses that needed replaced. Flushed entire system and replaced with new Porsche coolant. Used AirLIFT tool to remove air and check for leaks prior to refill. After running a few times I now have a small drip where the large water house connects (just under the coolant tank) to water pump. Assume O Ring is leaking or not fully seated. Can I drain just this line and correct seal and then just top off or do I risk getting air back in system ? If I drain coolant tank and line will the Airlift tool work here to avoid air in this line or what ? Not sure best procedure here. Thanks for any advice
