Is there a difference between sport and sport +
Is there a difference between driving in sport or sport plus on a pdk car if I drive the car in manual mode? I understand the difference when driving in automatic mode
Yes, definitely.
Sport Chrono + has faster shifts when in manual mode, both up and down. Down shifts are pretty agressive, as are upshifts (i.e. you will feel the jerk).
In auto mode, you would only want this for the track (arguably). It holds RPMs very high and shifts just as you would want the car to shift for track conditions [a majority of the time, at least]
I drive in manual mode 95% of the time, but I did use auto mode on the track after another PCA member told me, and I was impressed. However, it felt like I was driving an automatic on the track (which I was) and it felt disgusting

The computer is actually telling the clutches how fast/hard to shift the gears. There are actually 2 clutches in the PDK transmission. One clutch operates gears 1,3,5,7, and the other operates 2,4,6. The PDK transfers from one clutch to the other, which is why it is so fast. In essence, the other gear is already ready for you, both lower (for downshifting) and higher (for up-shifting). Here is a detailed explanation:
Normal mode is very tame, and it shifts slow as to not jerk the driver (the advantage is comfort). Sport mode tells the computer to shift faster, which is less delay, and causes the car to jerk. Obviously, this is much better for driving the car aggressive. Sport + is the fastest possible gear shift. Trust me, if you have sport chrono + you will notice a big difference.
